So I do a lot of board games, but have yet to venture into the RPG world. A FLGS has this for sale and a friend and I are considering giving it a try. Now if we do so, does that mean we would have one DM and one player only? Or is there a way we can both be players?

I know in most DnD there is a DM controlling things, I just wanted to know if it is different for this starter set as a way to introduce people to it.

I understand this is probably a basic and dumb question, but at the moment I am basic and dumb.

Often, a DM in a two player game will play a character in the party. It's a little extra work but not too difficult.

You can play that way, although I'm not sure how exciting or satisfying you'll find it. If you want a similar DM-less experience for the combat you can check out Dungeons & Dragons: Castle Ravenloft Board Game or Dungeons & Dragons: Wrath of Ashardalon Board Game. If you are interested in getting into the role playing more you should check out Roleplaying Games for two players.

I would suggest the board games as a better alternative for a 2-person game which offers replayability. You can play the starter adventure in the starter set (the choose-your-own-adventure style character creation thing) as a two player, GM-less adventure by just doubling the number of each creature you encounter. However, the set offers zero replayability and you'll end up with some odd difficulties because you're going to have to look up the alternative choices in the adventure in order to make 2 characters of different classes. Overall, it's likely to be a very unsatisfying experience. Having played both (the SS with my 8-year-old, the board games with both my son and adult friends), the board game is a much more enjoyable experience.
